Fecal sampling

The fish is anesthetized and the feces is carefully scraped out into the test tube (see video). The fish do not need to be euthanized, and the procedure can be done by fish farmers in connection with lice counting, for example. The samples are weighed at
reception, and therefore it does not matter if the sample volume varies. A sample volume of 0.2 ml of feces is sufficient.
It is sufficient to analyze 10 fish per cage (or tank) to get a good picture of the stress level and detect the beginning of sexual maturation. If the fish are monitored weekly, 6-8 fish per sample are sufficient. After sampling, the tubes are frozen at the usual -18°C before sending.
Plasma sampling
Blood samples should only be taken by authorized fish health personnel.
We recommend using Vacuette Lithium-Heparin tubes and associated cannulas and tube holders, but it is also possible to use EDTA vacuum tubes or serum tubes. 4 ml vacuum tubes work well for all fish sizes.
The fish is given an overdose of anesthetic before sampling. In fish over 100 grams it is easy to insert the cannula laterally in line with the posterior edge of the anal fin (Figure 1). In small fish it may be easier to insert the cannula ventrally, just behind the anal fin (Figure 2). Try to hit the tail vein, which is just below the spine. If necessary, adjust the cannula slightly up and down until the blood flows into the vacuum tube.
For our analyses, a sample volume of at least 150 µL of plasma is required.
Immediately after sampling, it is very important to turn the vacuum tube upside down several times to bring the anticoagulant coated on the inside of the glass into contact with the sample. Most manufacturers recommend turning the tube at least 6-8 times. Subsequently, the tubes are centrifuged at 2000 g for 10 minutes (or 3000 g for 5 minutes). Finally, the plasma is transferred to eppendorf tubes and frozen in a regular -18°C freezer before sending.

Submission of samples
Ensure that all samples are clearly labeled (either on the tubes themselves or on the bags). Package the samples in a
Styrofoam box and attach some frozen freezer elements so that the samples are kept cool during transport.
Drop off the package at the nearest post office or in-store mail, and send with the Bring concept "Express
next day". We can help with pre-filled return labels that can be stuck directly on the box. You will
then be invoiced for actual shipping costs afterwards.
